Heterogeneity of Human Whole Blood Platelet

چکیده
A quantitative high yield method utilizing isosmotic arabino-galactan (Stractan) solutions and isopycnic centrifugation was developed to isolate and to fractionate total human platelet populations into density-dependent subpopulations. Isolated platelets were free of factor VIll/von Willebrand factor and other plasma proteins. They responded to ADP, epinephrine, and collagen with a sensitivity equal to platelet-rich plasma platelets. The correlation of platelet density with volume and ultrastructure was examined for normal subjects. Recovery of total platelet populations averaged 92.76% ± 3.64% (SD). Normal individuals exhibited a narrow range of platelet buoyant density distribution. Computerized probability plot analysis f platelet volume distribution for 15 normal subjects’ total platelet populations showed a mean volume of 5.17 ± 0.46 cu j m (SD). Platelets with buoyant density 1.062 g/ml had a mean volume of 4.50 ± 0.48 cu jzm, while platelets with buoyant density > 1.071 g/ml, but 1.084 g/ml, had a mean volume of 5.32 ± 0.63 cu zm (SD). The volume difference by paired t test was significant, p > 0.001. Thin-section electron microscopy demonstrated a significant reduction of granule content in light platelets, as compared to heavy platelets, but an equal number of mitochondria for both groups. These differences of platelet volume and structure between light and heavy platelets suggested that aging may be a determinant of platelet heterogeneity.

Conversion from Platelet-Rich Plasma Platelet Production to Buffy Coat Platelet Component Production: Benefits and Limitations
Blood transfusion centers are under considerable pressure to produce platelet concentrates with high quality and safety due to the short shelf life of 3-5 days as well as possible bacterial and viral contaminations. For a variety of reasons, many Europeans have changed their methods of component production from whole blood using the North American standard platelet-rich plasma method to the buf...
متن کاملComparative Study of Four Platelet-Rich Plasma Methods for Preparing Platelet Concentrates
Background: Platelet preparations have been used for a wide variety of clinical applications such as hemorrhage, tissue engineering and cellular therapy. Platelet concentrates can be prepared by the apheresis method or from the whole blood using the Buffy-coat or Platelet-rich plasma methods. The purpose of this study was to compare four variations of platelet-rich plasma method based on double...
متن کاملThe Role of Human Adult Peripheral and Umbilical Cord Blood Platelet-Rich Plasma on Proliferation and Migration of Human Skin Fibroblasts
BACKGROUND Wound healing is a complex and dynamic process following damage in tissue structures. Due to extensive skin damage caused by burn injuries, this study determined the role of human adult peripheral and umbilical cord blood platelet-rich plasma on proliferation and migration in human skin fibroblasts. METHODS Platelet-rich plasma (5, 10, 15, 20 and 50% PRP) from human umbilica...
متن کاملPotatoes: Source of Laboratory Reagent for Human Platelets Research
Irish potatoes is rich in carbohydrate, with varied glucose levels. It is also a very good source of apyrase, an enzyme used for platelet storage. The aim of this study was to explore Irish potatoes as an easily available source for apyrase extraction and purification. Apyrase was extracted from 5 Kg of Irish potatoes. Platelets were derived from 5-10 ml of human whole blood and G6PD activity w...

Abstract Background and Objectives Human platelet antigens (HPAs) are polymorphic structures located on the membrane of platelets (PLTs). These antigens have important role in clinical situations like refractoriness to platelet transfusion which is the most important cause of transfusion related mortality and morbidity in patients by recurrent platelet transfusion. Alloimmunization against the...

Background: Human platelet antigens (HPAs) are part of platelet GP complexes have the potential to contribute to the autoantibody production. Moreover, these antigens demonstrate different patterns of distribution on different ethnic groups and variation in some types of diseases. This study was objected to determine the incidence of HPA-1 to -5 and -15 polymorphisms in the Iranians suffering f...
